We present prenatal diagnosis, molecular cytogenetic characterization and genetic counseling of a chromosome 15q24 microdeletion of paternal origin.A 34-year-old primigravid woman underwent amniocentesis at 17 weeks of gestation because of advanced maternal age. Amniocentesis revealed a karyotype of 46,XY.

Six distinct actin isotypes have been identified in mammalian cells. Description: Beta-actin (human gene and protein abbreviation ACTB/ACTB) is one of six different actin isoforms which have been identified in humans. This is one of the two nonmuscle cytoskeletal actins. Actins are highly conserved proteins that are involved in cell motility, structure and integrity. Alpha actins are a major constituent of the contractile apparatus. Beta-actin has been shown to interact with SPTBN2. In addition, RNA-binding protein Sam68 was found to interact with the mRNA encoding β-actin, which regulates the synaptic formation of the dendritic spines with its cytoskeletal components. Beta-actin has been shown to activate eNOS, thereby increasing NO production. An eight-amino acid residue (326-333) in actin has been shown to mediate the interaction between actin and eNOS. Recurrent mutations in this gene have been associated to cases of diffuse large B-cell lymphoma. Beta actin is usually used as a loading control, for among others, the integrity of cells, protein degradation, in PCR and Western blotting. Its molecular weight is approximately 42 kDa. |

Description: Beta actin is a protein encoded by the ACTB gene which is approximately 41,7 kDa. Beta actin is localised to the cytoplasm. It is involved in pathways such as blood-brain barrier and immune cell transmigration, regulation of actin cytoskeleton by Rho GTPases and development Slit-Robo signalling. This protein falls under the actin family. There are 3 main groups of actin isoforms in vertebrates namely, alpha, beta and gamma actins. Beta actin is a highly conserved proteins that is involved in various types of internal cell motility and are the major constituent of microfilaments. Beta actin is ubiquitously expressed in all eukaryotic cells. Baraitser-Winter syndrome 1 is another disease that can occur due to mutations. Simultaneous array comparative genomic hybridization (aCGH) analysis on amniotic fluid revealed a de novo 2.571-Mb microdeletion of 15q24.1-q24.2. Prenatal ultrasound findings were unremarkable except persistent left superior vena cava and enlarged coronary sinus.
The woman requested repeat amniocentesis at 22 weeks of gestation, and aCGH analysis confirmed the result of arr 15q24.1q24.2 (72,963,970-75,535,330) × 1.0 [GRCh37 (hg19)] and a 15q24 microdeletion encompassing the genes of STRA6, CYP11A1, SEMA7A, ARID3B, CYP1A1, CYP1A2, CSK and CPLX3.
